CJC-1295 no DAC, also called Modified GRF 1-29, is a synthetic analogue of GRF(1-29), the functional fragment of growth hormone-releasing hormone that sermorelin also represents. It incorporates four amino acid substitutions (D-Ala2, Gln8, Ala15, Leu27) that reduce susceptibility to the enzymatic degradation seen in native GHRH(1-29). Unlike CJC-1295 with DAC, it lacks the maleimide-based Drug Affinity Complex that covalently binds serum albumin, so its reported half-life is markedly shorter (on the order of 30 minutes) with a more pulsatile signalling profile. How does CJC-1295 no DAC differ structurally from CJC-1295 with DAC? Both share the same four substitutions on the GRF(1-29) backbone. The DAC form adds a Lys30 residue bearing a maleimidopropionyl group that binds albumin. The no-DAC form omits that addition entirely, which is the only structural difference between the two molecules.
